The effect of micelles on the steady-state and time-resolved fluorescence of indole, 1-methylindole and 3-methylindole in aqueous media.

Solutions of indole, 1-methylindole (1-MI) and 3-methylindole (3-MI) were prepared containing the surfactant Brij-35. Steady-state fluorescence measurements were performed with a Spex Fluoromax , with excitation at 280 nm and emission monitored over the range 300-500 nm. There was a blue shift in the emission as the concentration of surfactant rose. Time resolved spectra revealed shorter average lifetimes for 3-MI as the concentration of Brij-35 rose. At the highest concentration of Brij-35 used, 93% of the 3-MI molecules partition into the micelle. The equilibrium constant for association between the micelles and the 3-MI is 26 000/ M . Samples for studying the effect of lecithin on the vapour of 3-MI were prepared by placing 0. 5 ml 98 small mu M -3-MI in ethanol in a sealed cuvette and adding lecithin (0, 12. 5, 25 or 37. 5 small mu g). The solvent was then evaporated under Ar. The dry surfactant lecithin caused a reduction of 3-MI in the vapour phase.

Rapid and direct determination of wastewater bod values using a fluorescence technique.

Waste water was treated with 0. 5 mg/l allylthiourea to prevent nitrification and was subjected to BOD testing. Fluorescence analysis was performed on a spectrofluorimeter ( Fluoromax , SPEX instruments SA) where the monochromators were equipped with a single grating. The fluorescence signals were collected at 90 degree C from the incident-beam direction, with similar 10 scans/spectrum, with 1 nm wavelength resolution. The samples were gently stirred in high-grade quartz cuvettes to simulate turbidity. The measurement was performed at 350 nm (excitation at 280 nm). A linear relationship was established between the absorbance and the BOD (graphs presented) at 340 nm (excitation at 280 nm), and the correlation coefficients were calculated. The industrial implementation of the method will enable online processing and cost-effective control.
